This test measures only orthophosphate. Samples must be decomposed before total phosphate can be measured.

In a solution acidified with sulfuric acid orthophosphate ions (PO43–) and molybdate ions form molybdophosphoric acid. This is reduced to phosphomolybdenum blue (PMB), the concentration of which is determined reflectometrically.
